Assay Conditions

Reaction done according to ACE2:SARS-CoV-2 Spike S1 Inhibitor Screening Assay Kit (BPS Bioscience #79945) protocol. Buffer used is PBS + 0.1% BSA. Blocking buffer is Superblock PBS + 0.05% Tween-20. Coat protein (ACE2, His-tag) was added overnight at 4° 50 ng/well. Various amounts of biotinylated ligand (Spike S1[Biotin-labeled]) (0-400 ng) added for 1h at room temperature. Strep-HRP added for 1h at room temperature. Luminescence developed with ELISA ECL substrates.

Instructions for Use

Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.

Applications

Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
