Natural Killer (NK) cells are lymphocytes involved in the innate immune system that respond to infected cells and tumor cells, limiting these pathologies. They are now considered potential tools for adoptive cell therapies.
Although NK cells share a common lymphoid lineage with T and B cells, they do not require prior sensitization to specific antigens and recognize and eliminate targets cells independently. They operate through a delicate balance of activating and inhibitory signals, engaging in a vigilant surveillance of the body's cells. Upon triggering of activating signals or a lack of inhibitory signals, NK cells exert their effects by releasing cytotoxic granules, expressing death ligands, driving antibody-dependent cell cytotoxicity, and producing cytokines, thereby playing a pivotal role in the early stages of immune defense and immune regulation.
The therapeutic potential of NK cells has garnered considerable attention and researchers are exploring various avenues to harness their power for clinical applications. Adoptive NK cell therapy involves isolating, expanding, and infusing ex vivo activated NK cells into patients, offering a promising approach for treating certain types of cancers. Additionally, engineering NK cells to enhance their cytotoxicity and persistence, particularly using chimeric antigen receptors (CARs), is an active area of investigation. NK cells can also be utilized in the context of hematopoietic stem cell transplantation to improve outcomes and reduce complications. Moreover, ongoing research is uncovering ways to modulate NK cell activity pharmacologically, opening avenues for the development of novel immunotherapies.
