Colorimetric Assay Kits

Colorimetric plate-based assays are a simple method to screen for inhibitors of known protein interactions or protein activity. The general principle is similar to an ELISA in which one protein partner is coated onto a plate surface followed by incubation with the second protein. This is followed by a secondary reagent, such as streptavidin or antibody, conjugated to Horseradish Peroxidase (HRP) that detects the second binding partner. Unbound components are washed away after each step. The final readout is based on the overall activity of HRP in the well, which, in the presence of colorimetric substrate, increases the optical density of the well.  Individual kits will vary in design of components.
