Storage/Stability

At least 6 months at -80°C. This protein is highly sensitive to freeze/thaw cycles.

Assay Conditions

The ELISA was performed by coating 100 µl of ACE2-Fc at 2 µg/ml in PBS at 4˚C overnight. The plate was washed in PBS and blocked with 200 µl of 1% BSA in PBS for 1 hour at room temperature. Increasing amounts of Spike Trimer CH.1.1 were added to the plate (serially diluted in blocking buffer). The plate was washed in PBS before adding 100 µl of AffiniPure Peroxidase-conjugated Anti-His-Tag antibody (Jackson Immuno Research #300-035-240) diluted 1:5,000 in blocking buffer for 1 hour at room temperature. The plate was washed with PBS and 100 µl of MB/E Ultra-Sensitiv HRP Substrate (EMD Millipore #ES022-500ml) was added for 5 minutes at room temperature. The reaction was stopped with 100 µl of 0.6N H₂SO₄ and read at 450 nm.

Instructions for Use

Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.

Applications

Screen or titrate anti-spike antibodies or inhibitors; use as antigen control in assay design and optimization.
