Background

PAK4 is a recently identified member of the p21-activated kinases (PAKs) which have been implicated in the regulation of cell morphology, motility and transformation. These serine/threonine kinases are activated by and are effectors of small GTPases, cdc 42 and Rac. PAK4 belongs to the Group II PAKs which also includes PAK5 and PAK6. PAK4 has been shown to regulate cell morphology and cytoskeletal organization in mammalian cells.

Storage/Stability

Assay Conditions

Analysis of enzymatic activity was performed according to the Zlyte assay protocol (Invitrogen):
1. Different concentrations of PAK4 were incubated in a buffer containing 50 mM HEPES pH 7.5, 10 mM MgCl₂, 1 mM EGTA, 200 μM ATP, 0.01% Brij-35, and 2 μM substrate (SER/THR 14, Invitrogen) at room temperature for 1 hour.
2. Developer solution was added to the reaction and the reaction was stopped after 1 hour of incubation at room temperature.
3. Fluorescence was then detected using λexc=460±40 nm and λem=528±20 nm filters.

Instructions for Use

Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.

Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
