Description

Complex of human PI3 kinase (phosphatidylinositol-4,5-bisphosphate 3-kinase) catalytic subunit p110α (also known as PIK3CA) and regulatory subunit p85α (also known as PIK3R1).
Recombinant human p110α, encompassing amino acids 2-1068(end), contains an N-terminal Flag-tag and mutation of interest H1047R. Recombinant human p85α, full length encompassing amino acids 1-724 (end), does not contain a tag. The proteins were co-expressed in sf9 insect cells and formed a complex which was affinity purified.

Assay Conditions

25 μl kinase reaction is conducted in 40 mM Tris-HCl (pH 7.5), 10 mM MgCl₂, 100 μM ATP, 200 μM of PI3K substrate (BPS Bioscience #40560) and the PI3K enzyme for 18 minutes at 30°C. Conversion of ATP to ADP is detected using ADP-Glo® Luminescent Kinase Assay reagents (Promega Corp., Madison, WI).

Instructions for Use

Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.

Applications

Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
