Background

PTPRF or LAR is a member of the protein tyrosine phosphatase family with an extracellular region, a single transmembrane region, and two tandem intracytoplasmic catalytic domains. PTPRF has been shown to function in the regulation of epithelial cell-cell contacts at adherents junctions as well as in the control of beta-catenin signaling. An increased expression level of this protein was found in the insulin-responsive tissue of obese, insulin-resistant individuals, and may contribute to the pathogenesis of insulin resistance.

Storage/Stability

At least 6 months at -80°C.

Assay Conditions

20 mM HEPES, pH 7.2, 100 mM NaCl, 2 mM EDTA, 1 mM DTT, 1 mM pNPP and 160 ng/µl LAR. Assay was done at 30&Deg;C for 5 minutes, and the reaction was monitored at 405 nm continuously. Specific activity was calculated based on ε _pNPP= 18,000 M^-1cm^-1.

Instructions for Use

Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.

Applications

Useful for the study of enzyme kinetics and regulation, to dephosphorylate target substrates and for screening inhibitors.
