Background

CHK2 (checkpoint kinase 2, also known as CHEK2) is an evolutionarily conserved serine/threonine protein kinase that operates as a cell cycle checkpoint in response to DNA damage. Thus, it is rapidly phosphorylated and activated upon DNA damage. It is known to phosphorylate and inactivate phosphatase Cdc25C, thereby preventing entry into mitosis. CHK2 also stabilizes tumor suppressor TP53 and is a key player in the ATM DNA damage response and repair pathway. CHK2 mutations are observed in Li-Fraumeni syndrome, an inherited predisposition to cancer. Mutations in CHK2 are also linked to various spontaneous types of cancer.
