Description

Recombinant virus HCV1b (Hepatitis C Virus, genotype 1b), encompassing amino acids 3-181 and cofactor NS4A encompassing amino acids 21-32. This construct is a fusion protein corresponding to the serine protease NS3. The protein contains the mutation of interest (R155K). The construct contains an N-terminal Flag-tag followed by a His-tag (6xHis). The recombinant protein was affinity purified.

Assay Conditions

Assay was performed in 50 µl HCV assay buffer containing 50 mM Tris, pH 7.4, 150 mM NaCl, 5 mM DTT, 10% glycerol, and 5 µM fluorescently labeled HCV substrate peptide (Anaspec #AS-22991). After 10-minute incubation at room temperature, reaction was at ex350/em500.

Instructions for Use

Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.

Applications

Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
