Description

Recombinant human SYK (spleen associated tyrosine kinase), encompassing amino acids 359-635(end). This construct contains an N-terminal GST-tag. A short peptide linker placed between GST and the protein includes a Thrombin Cleavage Site to facilitate removal of the GST portion. The recombinant protein was affinity purified.

Storage/Stability

At least 6 months at –80°C.

Assay Conditions

Assay was done in Kinase Buffer 1 (BPS Bioscience #79334) using, 0.2 mg/ml Poly(Glu:Tyr) as a substrate with 20 µM ATP incubated at 30°C for 35 min. SYK, Catalytic, GST-Tag Recombinant (BPS Bioscience #40487) was titrated at various concentrations. The amount of ATP transferred was calculated using Kinase Glo max reagent (Promega).

Instructions for Use

Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.

Applications

Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
