Assay Conditions

BRD4 protein is incubated with biotinylated acetyl-histone peptide substrate 2.5 μM stock (BPS Bioscience #33000) in BRD assay buffer (BPS Bioscience #33001) in a 10 μl reaction for 1 hour at room temperature. AlphaLISA^® Nickle acceptor beads (PerkinElmer) are added, followed by Alphascreen^® Streptavidin-conjugated donor beads (PerkinElmer). After 1 hour of incubation at room temperature, Alphacounts are measured.

Instructions for Use

Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.

Applications

Useful for the study of bromodomain binding assays, screening inhibitors, and selectivity profiling.
