Assay Conditions

Assay was performed using BRD4 Inhibitor Screening Assay Kit, Cat. #32524. BRD4 protein is incubated with biotinylated acetyl-histone peptide substrate (Cat. #33000) in BRD assay buffer (Cat. #33001) in a 10 µl reaction for 1 hour at RT. AlphaLISA^® GSH acceptor beads (PerkinElmer) are added, followed by Alphascreen^® Streptavidin-conjugated donor beads (PerkinElmer). After 15 minute incubation at RT, Alphacounts are measured.

Instructions for Use

Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.

Useful for the study of bromodomain binding assays, screening inhibitors, and selectivity profiling.
