This anti-CD8 antibody is a purified recombinant antibody, which is labeled with R-Phycoerythrin (PE). This antibody has been tested by flow cytometry for specific binding to human CD8 on the surface of PBMCs (BPS Bioscience #79059).

PE is an oligomeric protein complex (270 kDa) from red algae that exhibits intensely bright red-orange fluorescence with excitation peak at 566 nm and an emission peak at 574 nm. The complex consists of six heterodimers, α subunit (18 kDa) and β-subunit (20 kDa), and an additional γ-subunit (34 kDa).
Background
CD8 is a co-receptor for the TCR (T cell receptor) in T cells, binding to MHC (major histocompatibility complex) class I proteins. CD8 is a typical marker of cytotoxic T cells and is involved in signaling. CD8 has two isoforms, a and b. CD8b recruits Lck (lymphocyte-specific protein tyrosine kinase) to the TCR-CD3 complex, and Lck phosphorylates multiple proteins involved in activation of cytotoxic T lymphocytes. It is thus critical for the lysis of cancer cells.
Storage/Stability
At least 6 months at 4°C. Protect from prolonged exposure to light. DO NOT FREEZE. The antibody solution should be stored undiluted between 2°C and 8°C.
Assay Conditions
One million PBMC cells (BPS Bioscience #79059) were blocked and stained with 1 µg of anti-CD8 antibody, PE-labeled (BPS Bioscience #102011) or PE Human IgG1 Isotype Control (BioLegend #403503) for 30 minutes on ice, washed three times, and analyzed by flow cytometry. Non-lymphoid cells were excluded from the analysis based on side scatter and forward scatter signals. Y-axis represents the cell count while X-axis indicates PE intensity.
Instructions for Use
For flow cytometry staining, the suggested use of this reagent is 5 µl (1 µg antibody) per million cells in 100 µl staining volume.
Applications
Useful for studying expression of CD8 on the cell surface by flow cytometry.
