The RdRp (RNA dependent RNA polymerase) complex is a complex of the three SARS-CoV-2 proteins NSP7, NSP8 and RNA polymerase/NSP12, reconstituted from the affinity-purified recombinant proteins. Recombinant SARS-CoV-2 RNA polymerase/NSP12, full length encompassing amino acids 1-932 (end), contains an N-terminal FLAG-tag. Recombinant SARS-CoV-2 NSP7, full length encompassing amino acids 1-84 (end), contains a C-terminal His-tag (6xHis). Recombinant SARS-CoV-2 NSP8, full length encompassing amino acids 1-198 (end), also contains a C-terminal His-tag (6xHis).
RNA-dependent RNA polymerase was incubated with FRET Dig-RNA Duplex in the presence of RNaseOUT and biotinylated ATP for 80 min at 37°C in a 10-ul reaction. Eu-labeled a-Dig antibody and Streptavidin-acceptor dye was then added followed by TR-FRET detection (317 => 620 nm, and 317 => 665 nm). Increase in TR-FRET is proportional to the amount of ATP incorporated in the RNA
