This human anti-PSMA antibody is a purified recombinant antibody, which is labeled with fluorescein isothiocyanate (FITC). This antibody has been tested by flow cytometry for binding to human PSMA on the surface of the PSMA CHO Recombinant Cell Line (BPS Bioscience #79461).

Background
PSMA (prostate-specific membrane antigen), also known as Folate hydrolase 1 or FOLH1, is an enzyme with folate hydrolase and peptidase activity. It is highly expressed in prostate cancer cells and is used as a prognostic indicator of prostate cancer. PSMA is used in the clinic as a target for PET (positron emission tomography) imaging in the detection and progression of prostate cancer, but radioactive analogs are used for the treatment of prostate cancer. It is also expressed in other tumor types and in a few normal tissues. PSMA is under extensive investigation as target for CAR-T cells and bi-specific antibodies-based cancer therapies.
Storage/Stability
At least 6 months at 4°C. Protect from prolonged exposure to light. DO NOT FREEZE. The antibody solution should be stored undiluted between 2°C and 8°C.
Assay Conditions
One million PSMA CHO cells (BPS Bioscience #79461) were blocked and stained with 1 µg of anti-PSMA antibody, FITC-labeled (BPS Bioscience #101977) for 30 minutes on ice, washed three times, and analyzed by flow cytometry. PSMA CHO cells (green) were compared to parental CHO-K1 cells (blue). The Y-axis represents the cell count, while the X-axis shows FITC intensity.
Applications
Useful for studying expression of PSMA on the cell surface by flow cytometry. For flow cytometric staining, the suggested use for this reagent is 5 µl (1 µg antibody) per million cells in 100 µl of staining volume.
