Description
Recombinant human KRAS (GTPase), isoform a, encompassing amino acids 2-186 (end). In this construct the N-terminal end of the protein contains a His-tag (6xHis) followed by an optimal TEV protease target sequence that allows cleavage of the His-tag from the protein (TEV: tobacco etch virus cysteine protease #50308). The protein also contains cancer-associated mutation G12C. The protein was affinity purified and loaded with GppNHp, a non-hydrolyzable GTP analog. Unbound GppNHp was removed by spin column. Ready for use in KRAS-RAF binding studies or inhibitor assays.

Assay Conditions
KRAS (G12C) GppNHp-loaded was tested for binding to the RBD of Raf1. KRAS (G12C) GppNHp-loaded was compared with KRAS(G12C) GDP-loaded (BPS Bioscience #100640) for RBD-RAF1 (BPS Bioscience #100519) binding by Bio-layer Interferometry (GATOR, Probe Life) and AlphaScreening (PerkinElmer).
Instructions for Use
Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.
Applications
Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
