This anti-PVRIG (Poliovirus receptor related immunoglobulin domain containing) antibody is a purified, recombinant human monoclonal antibody which recognizes human PVRIG. This recombinant antibody has been tested for specific binding to purified recombinant human PVRIG in an ELISA binding assay.

Background
PVRIG, also known as Poliovirus receptor related immunoglobulin domain containing protein or CD112R, is a natural killer (NK) cell immune checkpoint receptor, and it is also found in CD8+ T cells. PVRIG works as an inhibitory receptor and recognizes CD112 (also known as poliovirus receptor-related 2, PVRL2 or nectin-2) as its ligand. This binding results in lower responses of NK and T cells towards cancer cells. In T cells it inhibits the nuclear factor of activated T cells (NFAT). Blocking the interaction between PVRIG and its ligand, for instance with an antibody directed towards PVRIG, represents a novel strategy in checkpoint blockade immunotherapy.

Assay Conditions
The antibody was validated by measuring its binding to a PVRIG antigen in ELISA. PVRIG protein (BPS Bioscience #101712) was coated onto a 96-well plate overnight at 4°C (50 µl/well at a concentration of 2 µg/ml in PBS). The plate was washed 3 times with Immuno Buffer 1 (BPS Bioscience #79311) and blocked using 100 µl of Blocking Buffer 2 (BPS Bioscience #79728) for 1 hour at Room Temperature (RT). After removing the blocking buffer, 50 µl/well of serially diluted CD112R (PVRIG), Fc-Fusion, Avi-Tag, Biotin-Labeled (Human) Recombinant (BPS Bioscience #79270) in Blocking Buffer 2, was added for 1 hour at RT. After 3 washes, the plate was incubated with Streptavidin-HRP (BPS Bioscience #79742), washed again, and colorimetric HRP substrate was added. The reaction was stopped, and absorbance was read at λ=450 nm. The Blank value was subtracted from all values.
Instructions for Use
Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.
Applications
Useful for binding and neutralization of PVRIG in ELISA and in cellular assays.
