PARG is the major enzyme responsible for the catabolism of poly(ADP-ribose), a reversible covalent-modifier of chromosomal proteins. It reverses PARP-mediated ADP-ribosylation and plays a role in DNA damage responses. PARG inhibition may prove beneficial in cancer therapy.
