The eGFP/Firefly Luciferase RS4;11 Cell Line are engineered human lymphoblast RS4;11 cells expressing firefly luciferase and enhanced GFP (eGFP) driven by an EF1a promoter. The cells were generated by transduction with Firefly Luciferase-eGFP Lentivirus (BPS Bioscience #78741), which is a SIN (self-inactivating) lentivirus.

The cell line has been screened to confirm the absence of Mycoplasma species.
RS4;11 is a lymphoblast cell line derived from the marrow of a 32-year-old white female patient with acute lympholastic leukemia (ALL). This cell line has a KMT2A (histone-lysine N-methyltransferase 2A) rearrangement, which despite its low frequency leads to acute leukemia. It expresses HLA-C alleles, that bind the most expressed KIRs (killer-cell immunoglobulin-like receptors) and it is resistant to NK attack. Its characteristics make it an ALL-cell model and an appropriate negative control in NK cytotoxicity assays. The presence of eGFP and luciferase allows for easy assay readouts, making this cell line a convenient choice.
