Background
WNK4 or WNK lysine deficient protein kinase 4 is a member of the WNK family of cytosolic serine-threonine protein kinases and is part of the tight junction complex in kidney cells. WNK4 can regulate the balance between NaCl reabsorption and K(+) secretion. WNK4 negatively regulates the surface expression of SLC12A3, and the loss of this regulation may cause an inherited form of hypertension. WNK4 regulates the activities of several types of ion channels, cotransporters, and exchangers involved in electrolyte flux in epithelial cells. WNK4 is a positive regulator for the WNK-OSR1/SPAK-NCC cascade and is a potential target of antihypertensive drugs.

Assay Conditions
10 μl of varying amounts of WNK4 was mixed with 10 μl of 0.5 mg/ml PKA protein substrate. Reaction was initiated by adding 5 μl of 250 μM [33P]-ATP and incubating for 15 min at 30°C followed by spotting the reaction on phosphocellulose paper, fixing in 1% phosphoric acid, and assaying with a scintillation counter.
Instructions for Use
Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.
