Background
WNK2 (with no lysine kinase 2) is a cytosolic serine/threonine kinase of the WNK protein family that also include WNK1, 3 and 4. The WNK kinases feature a cysteine residue in place of the conserved lysine residue present in the ATP binding subdomain II of most kinases. These kinases control salt homeostasis and blood pressure. WNK2 is predominantly expressed in the brain, heart muscle, small intestine, and colon. WNK2 is involved in the modulation of growth factor-induced cancer cell proliferation through the MEK1/ERK1/2 pathway. WNK2 expression was found down-regulated epigenetically by promoter methylation in human gliomas.

Assay Conditions
10 μl of varying amounts of WNK2 was mixed with 10 μl of 0.5 mg/ml PKA protein substrate. Reaction was initiated by adding 5 μl of 250 μM [33P]-ATP and incubating for 15 min at 30°C followed by spotting the reaction on phosphocellulose paper, fixing in 1% phosphoric acid, and assaying with a scintillation counter.
Instructions for Use
Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.
