Background

Praja1 (along with Praja2, 53% identity) is a RING-H2 domain E3 (ubiquitin protein ligase) that is involved in the conjugation of ubiquitin to target substrates along with E1 and E2 enzymes. Substrates for Praja1 include Dlxin-1 (MAGE-D1, human) which regulates Dlx5 dependent transcriptional function (involved in osteoblast differentiation) and ELF and Smad3 involved in the TGF-β signaling pathway implicated in gastrointestinal cancers.
