Background

FGF19, or fibroblast growth factor 19, is part of the FGF growth factor family. It is involved in bile acid synthesis, where it functions as a hormone to regulate glucose and lipid metabolism. Defective levels of FGF19 can result in primary bile acid diarrhea, and FGF19 can be found in the liver of patients suffering from cholestasis. FGF19 overexpression has been found in cancer, such as prostate, bladder and lung cancer. Of note is its presence in almost half of the hepatocellular carcinomas. FGF19 is the ligand for FGFR4 (fibroblast growth factor receptor 4), and results in the activation of the receptor, acting like an oncogene. The development of specific inhibitors for FGFR4 activation by FGF19 may reveal as useful therapeutical tools for cancer treatment.
