Background

NUAK2 (NUAK family SNF1-like kinase 2), also known as SNF1/AMP kinase-related kinase (SNARK), is a member of the AMP-activated protein kinase family. It participates in cell-cell interactions and cell motility by regulating the conversion of F-actin to G-actin, contributes to neural development, cell proliferation and apoptosis. Loss of function mutations in NUAK2 can result in anencephaly, due to abnormal Hippo signaling pathway cytoskeletal regulation during neural tube closure. NUAK2 has also been involved in cancer development and progression, and it is overexpressed in gastric and cervical cancer and in glioma. Knockdown of NUAK2 in melanoma and cervical cancer cells, in vitro, inhibits cell proliferation and migration. Further studies and a deeper understanding of NUAK2 role and partners will help identify novel cancer therapy strategies.
