Background

SMARCA2 (SWI/SNF related, matrix associated, actin dependent regulator of chromatin, subfamily a, member 2) is a member of the helicase family of proteins, involved in regulating the transcription of certain genes and their chromatin structure, DNA repair and genome instability. SMARCA2 genes are often silenced in tumors, resulting in low expression of this protein in most cases. High expression of SMARCA2 has been linked to a good prognosis and to benign differentiated tumors. The selective targeting of SMARCA2 versus SMARCA4 has been a challenge. The use of PROTAC-mediated degradation has proved an alternative for specific degradation of SMARCA2.
