Background

PKM1 and PKM2 are different splicing products of the PKM gene (exon 9 for PKM1 and exon 10 for PKM2). The M1 isoform (PKM1) is expressed in muscle and brain, and the M2 isoform is expressed during embryogenesis, in adipose tissue and pancreatic islets, and is the predominant form found in cancer cells.

Storage/Stability

Assay Conditions

Assay was done in buffer containing 50 mM Tris (pH 7.4), 10 mM MgCl₂, 50 mM KCl, 2 mM ADP, 10 mM phosphoenolpyruvate (PEP) and PKM2 (BPS Bioscience #50295) titrated at various concentrations. Reactions were done in 50 µl duplicates and incubated at room temperature for 15 minutes. ATP production was detected using Kinase-Glo® Max (Promega).

Instructions for Use

Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.

Applications

Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
