Assay Conditions

MAT2A #71401 was tested according to the MAT2A Inhibitor Screening Assay Kit catalog #71402 with enzyme tested from 0.9375 ng/µl-120 ng/µl. The reactions were conducted at room temperature for 45 minutes in a 25 µl mixture containing 50 mM Tris-HCl, pH 8.0, 10 mM MgCl2, 50 mM KCl, 0.1 mg/ml BSA, various amount of MAT2a enzyme, 200 μM LMethionine, and 200 μM ATP. After the enzymatic reactions, 50 μl of colorimetric detection reagent was added to each well and the reactions were incubated for another 15 min before the absorbance at 630 nm of each well was measured using a Tecan Infinite M1000 microplate reader.

Instructions for Use

Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.
