Background

PKLR is a pyruvate kinase which catalyzes the transphosphorylation of phohsphoenolpyruvate into pyruvate and ATP. This reaction is the rate-limiting step of glycolysis. PKLR gene encodes the L- and R-type isoenzymes through alternate splicing events controlled by different promoters. L-type isoform can also appear as a tetramer and is upregulated by glucose with implications in maturity-onset diabetes of the young.

Assay Conditions

A 50 µl reaction is conducted in a buffer containing 50 mM Tris (pH 7.4), 10 mM MgCl₂, 50 mM KCl, 2 mM ADP, 10 mM phosphoenolpyruvate (PEP) at room temperature for 15 min. ATP production is detected using Kinase-Glo® Luminescent Kinase Assay Platform.

Instructions for Use

Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.

Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
