Background

Phosphodiesterases (PDEs) play an important role in the dynamic regulation of the second messengers cAMP (cyclic adenosine monophosphate) and cGMP (cyclic guanosine monophosphate) signaling, by hydrolyzing them. The PDE superfamily is composed of 11 families, with PDE4, 7 and 8 being cAMP-specific hydrolases, and thus regulating positive and negative responses to it. PDE4 is a subfamily of PDEs that includes isoenzymes A to D, and that can be long, short or supershort, to a total of at least 20 different known non-redundant isoforms. PDE4 proteins are involved in cancer, depression, autism, and inflammation. Inhibitors of PDE4 are being used for the treatment of psoriasis, COPD (chronic obstructive pulmonary disease) and atopic dermatitis. PDE4A4 is a long isoform found in several tissues, including the pituitary. It has been linked to GH (growth hormone)-secreting adenomas. It can interact with AIP (AH receptor-interacting protein), a tumor suppressor. The development of PDE4A specific inhibitors and a deeper understanding of the role of each PDE4A isoform in health and disease will prove beneficial.
