Assay Conditions

BRD9 protein is incubated with biotinylated acetyl-histone peptide substrate BET Bromodomain Ligand in BRD9 assay buffer in a 10 µl reaction for 30 min at RT. AlphaLISA^® ® GSH acceptor beads (PerkinElmer) were then added, and the mixture was incubated for 30 minutes at room temperature (RT). Alphascreen^®® Streptavidin-conjugated donor beads (PerkinElmer) were then added. After 10 minute incubation at RT, Alphacounts are measured.

Instructions for Use

Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.

Useful for the study of bromodomain binding assays, screening inhibitors, and selectivity profiling.
