This anti-IL2RA (interleukin-2 receptor alpha subunit) antibody is a purified, recombinant human monoclonal which recognizes IL-2RA (also known as CD25). This affinity purified antibody has been tested for specific binding to the human recombinant protein IL2RA, Avi-His-Tag Recombinant (BPS Bioscience #101203) in a colorimetric ELISA binding assay.

Background
IL2RA (interleukin-2 receptor alpha chain), also known as CD25, is one of the subunits of the high affinity IL-2 receptor. IL-2 receptor is composed of IL2RA, IL2RB and IL2RG, and it is crucial for immune cell responses. Il-2 receptors with low and intermediate affinity also exist, controlling the sensitivity of specific cell types to IL-2. IL-2 binding to the IL2R activates downstream pathways, starting with phosphorylation of JAK1 (Janus tyrosine kinase) and JAK3. The understanding and manipulation of the IL-2/IL2R.

Assay Conditions
This antibody was validated by measuring Anti-IL-2RA binding to IL2RA antigen by ELISA. IL2RA, Avi-His-Tag Recombinant (BPS Bioscience #101203) was coated onto a 96-well plate overnight at 4°C (50 µl/well at a concentration of 2 µg/ml in PBS). The plate was washed 3 times with Immuno Buffer 1 (BPS Bioscience #79311) and blocked using 100 µl of Blocking Buffer 2 (BPS Bioscience #79728) for 1 hour at Room Temperature (RT). After removing the blocking buffer, serially diluted purified Anti-IL-2RA Neutralizing Antibody in Blocking Buffer 2 (50 µl/well) as added for 30 minutes at RT. After 3 more washes, the plate was incubated with Goat Anti-Human IgG Fc (HRP) (Abcam #ab97225), washed, and incubated with Colorimetric HRP substrate. The reaction was stopped, and absorbance was read at λ=450 nm. The Blank value was subtracted from all values.
Instructions for Use
Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.
Applications
Useful for studying the binding and neutralization of IL-2RA in ELISA and in cellular assays.
