T lymphocytes are white blood cells involved in the adaptive immune response. They are activated by infected, foreign, or cancerous cells. T cells help activate other immune cells and regulate the immune system, while developing immunological memory to enable a rapid response upon future exposure to the same pathogen.

The therapeutic applications of T cells hold great promise for improving the treatment of various diseases by harnessing and manipulating the power of the body's immune system. T cells have been exploited as therapeutic agents in several disease applications, particularly for cancer treatment and autoimmune disease management.
Some of the key therapeutic applications of T cells include adoptive cell therapy using T cells engineered to express a chimeric antigen receptor (CAR) or a TCR (T cell receptor) specifically directed against a cancer antigen, for the treatment of cancer or auto-immune diseases. Another approach is to use checkpoint inhibitors, which help unleash T cell responses by blocking the inhibitory signals that cancer cells use to evade the immune system. Multi-specific T cell engagers, which bind simultaneously to a specific cell surface protein on tumor cells and to a T cell-specific protein such as CD3, are being assessed for their capacity to recruit T cells to the tumor and activate and immune response.
T cells can also be utilized for non-cancer applications as well. Regulatory T cells (Tregs), which suppress excessive immune responses, have been explored for treating auto-immune or inflammation-related diseases. Furthermore, engineered T cells that can recognize and eliminate virus-infected cells have shown potential in treating persistent viral infections like HIV, cytomegalovirus (CMV), and hepatitis B and C.
T cell lymphocytes are mainly composed of two T cell subpopulations, αβ and γδ, which are distinguished by the expression of an αβ or a γδ TCR, respectively. The αβ cells are the predominant T cell subset in the circulation and recognize antigens presented by MHC (major histocompatibility complex) molecules. The γδ-T cell subset is much less abundant and recognize antigens independently of MHC presentation. Both can target and eliminate infected or abnormal cells. However, γδ-T cells injected into patients have a lower risk of causing graft-versus-host disease compared to αβ-T cells, while retaining cytotoxicity against a wide range of tumor types.
