BPS Bioscience offers fast and convenient PARPtrap™ screening services for the discovery and evaluation of lead compounds. Our unique PARPtrap™ assay can help determine cytotoxic potency of small molecule inhibitors by measurement of PARP1-DNA complex formation. The assay uses fluorescence polarization technology and a fluorescent-labeled nicked DNA. Without the PAR reaction, PARP1 binds to the nicked DNA, resulting in the emission of highly polarized light. However, after autoribosylation of PARP1, the nicked DNA dissociates from PARP1 and rotates freely, emitting less polarized light (Fig. 1). View Figure 1. PARPtrap™ Assay Schematic
