Description

Recombinant human protein kinase ABL1 (variant b) encompassing amino acids 248-531 that correspond to the kinase domain. In this construct the N-terminal end of the protein contains a His-tag (6xHis) followed by an optimal TEV protease target sequence that allows cleavage of the His-tag (TEV: tobacco etch virus cysteine protease BPS Bioscience #50308). The protein was affinity purified.

Assay Conditions

Assay was done in a kinase buffer containing 0.2 mM DTT using Abltide substrate (0.2 mg/ml) and 20 µM ATP. Reaction was done at 30°C for 40 min. Amount of ATP transferred was calculated using Kinase-Glo reagent (Promega)

Instructions for Use

Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.

Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
