Storage/Stability

At least 6 months at -80°C.

Assay Conditions

25 mM Tris/HCl, pH 8.0, 137 mM NaCl, 2.7 mM KCl, 1 mM MgCl₂, and 0.1 mg/ml BSA, 20 µM Fluorogenic HDAC/SIRT substrate 1 (BPS Bioscience #50032), 500 µM NAD+ and Sirtuin 3 (31.25 ng - 2000 ng). Incubation condition: 1 hour at 37°C followed by treatment with 50 µl of SIRT developer (BPS Bioscience #50089) for 15 min at room temperature. Fluorescence intensity is measured at ex360/em460.

Instructions for Use

Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.

Applications

Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
