Proteases are catalytic enzymes found in all forms of life, including viruses, that regulate a wide range of vital physiological processes. In animals, they are critical for digestion of food, lysosome and proteasome degradation, apoptosis, blood coagulation, cell cycle regulation, cell signaling, and more. The dysregulation of protease activity can lead to pathologies such as arthritis and inflammatory diseases, cardiovascular disorders, cancer, osteoporosis, Alzheimer’s, and other neurological disorders. Several small molecule protease inhibitors are already on the market and many more are in development.

Viral proteases, such as SARS-CoV-2 3CL Pro, are essential to viral infection and replication and are attractive targets for anti-viral therapeutics.
