Assay Conditions

Assay was performed with PDE Assay Buffer (BPS Bioscience #60393), 0.1 mg/ml BSA, 500 µM cAMP, 0.05 Ku/µl 5’ Nucelotidase (Enzo-BML-K1307), and PDE4B1, GST-Tag (BPS Bioscience #60041) serial diluted at various concentrations. The reaction was incubated at 37°C for 20 min. Following the reaction, malachite green reagent was added and then read 10 min after with a microplate reader capable of reading absorbance at 620 nm.

Instructions for Use

Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.

Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
