Description

Recombinant human PDE3B (phosphodiesterase 3B), encompassing amino acids 592-1112(end). This construct contains an N-terminal GST-tag. A short peptide linker placed between GST and the protein includes a Thrombin Cleavage Site to facilitate removal of the GST portion. The recombinant protein was affinity purified.

Assay Conditions

Assay was performed with 10 mM Tris-HCl, pH 7.4, 10 mM MgCl2, 0.05% Tween-20, 200 µM cGMP, 2.5 kU 5'-nucleotidase, 0.1 mg/ml BSA, and material serial diluted. The reaction was incubated at 37°C for 20 min. Following the reaction, malachite green reagent was added and then read 10 min after with a machine capable of reading absorbance at 620 nm.

Instructions for Use

Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.

Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
