ATP citrate-lyase is an important enzyme for cytosolic acetyl-CoA synthesis. During fatty acid synthesis, Citrate and CoA form acetyl-CoA and oxaloacetate.

ATP citrate-lyase has emerged as new target for cancer treatment. Studies have found that many types of cancer including glioblastoma, colorectal cancer, breast cancer, non-small cell lung cancer, and hepatocellular carcinoma experience abnormally high levels of ATP citrate-lyase.

Human ATP citrate lyase (ACLY) (GenBank Accession No. NM_001096), full length, with N-terminal GST-tag, MW=147 kDa, expressed in baculovirus expression system.

Specific Activity:
≥300 pmole/min/μg. 
Assay Conditions:
A 25 µl ACLY reaction is conducted in a buffer containing 40 mM Tris (pH 8.0), 10 mM MgCl2, 2 mM DTT, 2 mM potassium citrate, 0.1 mM CoA and 6-100 ng human ACLY at 30 oC for 60 min. Enzyme activity was assayed using ADP-Glo reagents (Promega)
45 mM Tris-HCl, pH 8.0, 124 mM NaCl, 2.4 mM KCl, 18 mM glutathione, 10% glycerol, and 3 mM DTT
