Substrate for ceramide kinase (CERK), optimized for use with β-Octylglucoside mixed micellar assay. Diluting ceramide 12.5 fold in ceramide diluent buffer is recommended.

Storage/Stability

At least 6 months at –20°C when stored as components. Prepare substrate immediately before use.

Assay Conditions

Ceramide Kinase (BPS Bioscience, #40617) reaction was performed for 25 minutes at 37°C using 100 μM CERK substrate. Kinase-Glo® (Promega) was used to detect the enzyme activity.

Applications

Useful as a substrate for CERK activity assays.

Notes

Substrate Preparation

Prepare a fresh 1:4 mixture of ceramide and ceramide buffer. Sonicate the mixture 3 times for 2 min. using a bath sonicator to make a 2.5 X solution (250 μM ceramide). A final ceramide concentration of 100 μM is recommended. Discard any unused substrate.

Warnings

Prepare substrate immediately before use. Avoid freeze/thaw cycles.
