Background

Dipeptidyl peptidase-7 (DPP7), also known as DPP2, is a serine aminopeptidase found in quiescent lymphocytes. Inhibition of this enzyme induces apoptosis, and levels of DPP7 are elevated in many tumors, suggesting that DPP7 may be involved in a novel pathway for tumor suppression.

Storage/Stability

>6 months at -80°C.

Assay Conditions

Test wells contained 0.01M Tris-HCl pH7.4, 0.01M MgCl2, 0.05% Tween-20, 5 µM DPP substrate 1, and various amounts of DPP7(0-200ng). Standard wells contained 0.01M Tris-HCl pH7.4, 0.01M MgCl2, 0.05% Tween-20, and various amounts of Fluorescent AMC standard (0.01 µM-1.25 µM). Incubated for 10 minutes at room temperature. Fluorescence intensity was measure at excitation 360 nm and emission 460 nm.

Instructions for Use

Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.

Applications

Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
