Assay Conditions:
25 μl ACC2 reaction is conducted in a buffer containing 30 mM HEPES (pH 7.4), 2 mM MgCl2, 2 mM potassium citrate, 1 mM DTT, 100 μM ATP, 12 mM NaHCO3 and 20 μM acetyl-CoA at 30°C for 45 min. ATP reduction is detected using ADP-Glo® Luminescent Assay Platform (Promega).
Formulation:
40 mM Tris-HCl pH 8.0, 110 mM NaCl, 2.2 mM KCl, 0.04% Tween-20, 3 mM DTT, 20% glycerol and 100 mM Imidazole.
